Abstract:
Since the number of patients suffering from the infectious disease melioidosis is rising worldwide and antibiotic treatment is complicated and not always successful, research regarding the environmental organism and human pathogen Burkholderia pseudomallei is highly important. Cell death mechanisms were shown previously to play a critical role during innate immune response against microbial infections. Most research regarding B. pseudomallei is based on murine models; in contrary, we use human monocyte-derived primary macrophages to present a more detailed picture in the human host response during infection. In this study, we (i) analysed the cytotoxicity of different B. pseudomallei wild type and mutant strains, (ii) compared cell death mechanisms occurring during B. pseudomallei and B. thailandensis infections, and (iii) investigated apoptotic and pyroptotic players during microbial infections.
