Abstract:
The gram-negative pathogen Burkholderia pseudomallei causes the infectious disease melioidosis. In the course of the innate immune response it leads to the activation of the inflammasome in macrophages which strongly contributes to bacterial killing. The non-canonical inflammasome is accompanied with sensing the lipopolysaccharide (LPS) and activation of the inflammatory caspases 4 and -5, which leads to pyroptosis, an inflammatory cell death. The noncanonical inflammasome in B. pseudomallei infected human macrophages is still understudied. The aim of this work was to determine the relevance of casp-4 as regulator of the non-canonical inflammasome and the contribution of the canonical inflammasome mediator casp-1 during late B. pseudomallei infection in the monocytic cell line THP1 and human monocytes derived macrophages.
