Abstract:
In situ sequencing is a cutting-edge technology that allows the direct sequencing of genetic material without losing the context of the whole tissue. This technique has tremendous potential in the field of cancer research, as it provides a high-resolution view of genetic changes that occur at the cellular level. Colorectal cancer is one of the most common and deadly forms of cancer, and a better understanding of the genetic changes that occur in this disease is crucial for the development of more effective treatments. In the study, in situ sequencing was performed to analyze the genetic changes in a series of colorectal cancer samples. A novel in situ sequencing technique was implemented on 10 CRC samples to identify genes which are responsible for cancer relapse. Additionally, to determine the genes associated with tumor spreading, various gene panels were applied to 23 colorectal cancer samples. In conclusion, this Master’s thesis provides insight into the feasibility and utility of in situ sequencing in the study of colorectal cancer and highlights the potential of this technique to contribute the development of new and more effective treatments for this devastating disease.
