Characterization of long non-coding RNAs in renal cell carcinoma.

Abstract:
Long non-coding RNAs (lncRNAs) are non-protein coding RNAs longer than 200 nucleotides (nts), that are known to execute several regulatory mechanisms like regulation of transcription, translation and protein interaction. It is suggested that lncRNAs play important roles in initiation and progression of cancer and thus may provide novel biomarkers or therapeutic targets. In this master thesis, we investigated the role of lncRNA LINC01111 in renal cell carcinoma based on preliminary data showing that this lncRNA is associated with tumor spheres. Survival analysis of clear cell renal cell carcinoma (ccRCC) patient data showed, that high expression levels of LINC01111 are associated with poor survival. Furthermore, malignant tissue has overall higher LINC01111 levels compared to healthy kidney tissue. Thus, LINC01111 could provide a novel biomarker for ccRCC detection and prognostic purposes. The impact of this lncRNA on hallmarks of cancer was further explored in vitro by investigation of proliferation, migration, invasion and angiogenesis. Unfortunately, we couldn’t identify significant changes in these hallmarks after silencing of LINC01111. LINC01111 was detected, due to its downregulation in RCC cell lines when grown in non-adherent 3D culture systems via micro-array analysis. Knockdown of LINC01111 caused significant down-regulation of the stem cell factor NANOG in RCC-FG. Thus, LINC01111 could be involved in maintaining stem cell properties of cancer cells inside the primary tumor mass.
