Biological role of miR-146a in renal cell carcinoma.

- MicroRNAs (miRNAs) are endogenous, small, non-coding RNAs that are involved in important processes for the development of cancer. MiRNAs might be considered as potential diagnostic biomarkers in renal cell carcinoma (RCC). Based on microarray analysis miR-146a-5p showed upregulated expression levels in clear cell lines and its influence on the ‘’Hallmarks of Cancer’’ was therefore closely investigated in this study. Transient gain and loss of function experiments were performed in different clear cell RCC cell lines to examine the impact of miR-146a-5p on cell proliferation, migration, invasion and angiogenesis. MiR-146a-5p gene was frequently amplified in clear cell RCC tumors and high levels of expression were correlated with poor patient prognosis. In this study, we furthermore showed that high miR-146a-5p expression led to reduced migration compared to control. Moreover, it could be demonstrated a positive effect on angiogenesis, resulting in enhanced vessel growth. Furthermore, we identified CXCL8 and JAG 1 as two potential target genes, which might be involved in the migration and angiogenesis process. In conclusion, miR-146a-5p positively influences angiogenesis in primary tumors and promotes tumor growth by nutrient and oxygen intake. In contrast, inhibition of miR-146a-5p might cause metastases due to enhanced migration, highlighting the potential role of this miRNA as a novel prognostic marker and in the development of new therapeutic strategies in RCC.
