Abstract:
BACKGROUND: Several gene polymorphisms, including complement factor H (CFH) Tyr402His, complement factor B (CFB) Leu9His and complement component 2 (C2) IVS10C > T, have previously been associated with altered risk for exudative age-related macular degeneration (AMD). A complement component 3 (C3) Arg102Gly gene polymorphism has been identified as a novel risk factor for exudative AMD in both a British and North American population. The present study was set to investigate a hypothesized association between the C3 102Gly allele with exudative AMD in an Austrian population. METHODS: Genotypes of the C3 Arg102Gly, CFB Leu9His, C2 IVS10C > T and CFH Tyr402His polymorphisms were determined in 269 patients with exudative AMD and 155 control subjects. RESULTS: Presence of the C3 102Gly allele was significantly associated with exudative AMD (p = 0.001). Univariate analysis revealed an odds ratio of 1.71 (95% confidence interval (CI): 1.13-2.60) for exudative AMD among heterozygotes for the C3 102Gly allele compared to those carrying the wildtype, while an OR of 3.76 (95% CI: 1.25-11.34) was observed among subjects carrying the homozygous C3 102Gly/Gly genotype. In a stepwise logistic regression model including CFH Tyr402His, CFB Leu9His and C2 IVS10C > T genoytpes, odds ratios of 1.88 (95% CI: 1.18-2.99) for heterozygous and 3.46 (95% CI: 1.07-11.16) for homozygous carriers of the C3 102Gly allele were observed. CONCLUSIONS: Our data provide further evidence that in addition to other complement gene polymorphisms the C3 102Gly variant is associated with exudative AMD in an Austrian population.
