Abstract:
Endothelial cells release factors that regulate dilatation and contraction of the vessels. They play an important role in modulating both the inflammatory response and vasomotor abnormalities that occur in coronary artery diseases. This endothelial function is associated with changes of intracellular Ca2+ concentration. For this study we used spatially and temporally resolved measurements of local C2+ - concentration in human endothelial cells cultured in high glucose containing medium. Deconvolution techniques procedure allowed determination of intracellular Ca2+ concentration and its distribution into cellular compartments. We also used a confocal microscope for visualization of intracellular compartments (endoplasmatic reticulum, mitochondria) under normal and pathological conditions. We showed that the interrupted connection between superficial compartments and membrane channels is already the beginning of the cell damage in diabetes.
