Abstract:
BACKGROUND: Retinal artery occlusion (RAO) is a common vision-threatening disease. Several risk factors including arterial hypertension, diabetes mellitus and atherosclerosis have been identified. Atherosclerosis itself is a chronic low-grade inflammatory disease with a distinct proinflammatory cytokine profile. Among others, tumor necrosis factor alpha (TNF-alpha), a proinflammatory cytokine, contributes to atherogenesis and two TNF-alpha gene polymorphisms (TNF-alpha -308G > A and -238G > A) have previously been shown to modulate TNF-alpha expression. Consequently, the present study was set to investigate a hypothesized association between the TNF-alpha -308G > A and -238G > A polymorphisms and the presence of RAO. MATERIAL AND METHODS: Genotypes of the TNF-alpha -308G > A and -238G > A polymorphisms were determined in 240 patients with RAO and 335 control subjects. Determination of genotypes was done using 5' exonuclease assays (TaqMan). Results: Genotype distributions and allele frequencies of both TNF-alpha polymorphisms did not significantly differ between patients with RAO and control subjects (p > 0.05). Prevalences of arterial hypertension, diabetes mellitus and ever-smoking status were significantly higher in patients than among controls (arterial hypertension: 77.1% vs. 52.2%, p < 0.001; diabetes mellitus: 13.8% vs. 6.3%, p=0.002; ever-smoking status: 55.0% vs. 23.6%, p < 0.001). CONCLUSIONS: Our data suggest that both investigated TNF-alpha polymorphisms are not associated with increased risk for RAO in a Central European population.
