From early identification to early intervention

Abstract

Rett Syndrom, eine schwerwiegende neurologische Erkrankung, durch de novo-Mutationen des MECP2 Gens verursacht, hat einen vierphasigen Verlauf: Nach einem mehr oder weniger unauffälligen ersten Lebensjahr (Phase 1) kommt es zur Regression (Phase 2), in der erworbene Fähigkeiten wie Sprache und motorische Funktionen verloren gehen. Mit Phase 3 tritt eine relative Stabilisierung ein; Phase 4 ist gekennzeichnet von erneuten motorischen und kognitiven Beeinträchtigungen. Da eine Diagnose erst im Kleinkind- bis Schulalter erfolgt, gibt es wenig systematisches Wissen über die Prä-Regressionsphase. Ziel des eingereichten Projekts ist die Erstellung spezifischer Entwicklungsprofile dieser ersten Phase und zwar in Abhängigkeit von Locus und Typ (missense/nonsense) der Mutatuion.
Unsere detaillierten Analysen der Genotyp-Phänotyp Beziehungen und des prä-diagnostischen Verlaufs basieren auf Audio- und Videoaufnahmen, die uns von betroffenen Patienten und deren Familien weltweit zur Verfügung gestellt werden.

Für neue Analysemöglichkeiten beantragen wir ein Upgrade des vorhandenen Systems Noldus The Observer XT. Mit erhöhter Analysekapazität und -genauigkeit kommen wir unserem längerfristigen Ziel der besseren Versorgung betroffenener Mädchen und ihrer Familien durch das verlässliche Identifizieren von Frühzeichen einen Schritt näher.
