High-Resolution Melting Real-time PCR for Promoter Methylation Analysis in Prostate Cancer

Abstract
Cancer represents a major health problem worldwide. Early detection of disease generally results in an improved clinical outcome for cancer patients. Thus, the search for biomarkers that can reliably show evidence of cancer before it metastasizes is still ongoing. Promoter hypermethylation analysis is a promising tool for cancer diagnostics and as a prognostic marker. This has been shown in prostate cancer and other cancers. Prostate cancer is the most commonly diagnosed cancer in men in Europe. Hence, it is a suitable model for the proposed analysis. The objective of the present proposal is to establish and evaluate the feasibility of High Resolution Melting Realtime PCR as a simple, cost-effective alternative to real-time methylation specific PCR for analyzing DNA methylation status in prostate cancer patients. This proposed novel technique may facilitate further research and clinical implementation of prognostically important cancer specific methylation analysis.
